Product Description

The pan anti-O-GlcNAc monoclonal antibody specifically recognizes O-linked glycoproteins. Besides common applications in Western blotting and immunoprecipitation, the antibody has been well utilized for proteomic screening to o-linked glycoproteins from protease-digested proteins both in prokaryotic and eukaryotic cells.?


Source/Purification

This product is produced by immunizing Balb/c mice with a special immunogen and the ascites was immunoaffinity purified by protein G.

Important

For Western blotting, incubate ?membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 for ?two hours at room temperature with gentle shaking.??

Western blot analysis?on 30 μg of extracts from HeLa whole cell with (lane “+”) or without (lane “-“) treatment of the inhibitor of o-acetylglucosaminidase PUGNAC (100 μM, 24hr) using anti-O-GlcNAc mouse mAb. The blot was reprobed with beta-tubulin antibody as a loading control.


Scientific Description

O-Linked β-N-acetylglucosamine modification (GlcNAcylation) is a glycosylation in which the monosaccharide β-N-acetylglucosamine (GlcNAc) attaches to serine/threonine residues via an O-linked glycosidic bond and is found mostly within the cytoplasm or nucleoplasm. GlcNAcylation regulates several important cellular processes, such as signal transduction, protein expression, degradation, embryonic stem cell pluripotency and trafficking. This post-translational modification is regulated by OGT and O-GlcNAcase (β-N-acetylglucosaminidase) enzymes, whereas OGT catalyzes the attachment and O-GlcNAcase catalyzes the removal of O-GlcNAc to proteins. O-GlcNAc modification has been observed on histones: H2A at T101, H2B at S36 and S112, H3 at S10 and T32, and H4 at S47.

Storage & Stability

The antibody is kept in PBS with 50% glycerol and 0.01% sodium azide. Upon receipt, please centrifuge the antibody at 12,000 x g for 20 seconds and store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Stable for 12 months from date of receipt. Leave the antibody at room temperature for 2 minutes and gently mixed using pipette before usage.
